High Resolution

dPC™ chips are precision manufactured to have a difference of 0.05 pH units between adjacent gel plugs. This gives proteomic scientists the ability to fractionate and concentrate their samples by isoelectric focusing, enabling separation of proteins or peptides often found in the same fraction by other technologies. Protein Forest's digital ProteomeChip™

1. Resolution up to 0.05 pH units
2. Know the pI of your protein or peptide
3. Increase proteome coverage
4. Separate highly abundant from low abundant species
5. Facilitates study of pI isoforms including post-translational modifications

Legend: The figure above shows the separation of 2 proteins Alpha 2-macroglobulin and PKDP1 by pI from plasma that was processed using the dPC™ platform.

Reproducibility

Automated manufacturing of dPC™ chips and the discrete nature of the gel features, assures that the same proteins or peptides will always be found in the same gel plugs from chip to chip and experiment to experiment.

1. Greater confidence in your data
2. Fewer replicates required
3. Eliminates variability associated with gel band cutting

System Reproducibility

Legend: High reproducibility observed using the dPC™ system. The technical replicates of the mass spectrometer are 79%. Any variability introduced by the dPC™ chip was determined by running multiple dPC™ fractions. The ratio provides the overall reproducibility of the dPC™ platform, 92%.

dPC™ Isoelectric Western Blot Kit iWB™

Western Blot Kit Coming Soon - Contact Protein Forest® for details
For antibody detection of dPC™ separated proteins, a semi-dry blotting device from Protein Forest® is used. The rigid format of the dPC™ allows direct transfer of proteins from the chip to a PVDF membrane while maintaining the digital array format. Protein Forest® will offer a complete kit with optimized reagents for electroelution of proteins from the dPC™.

dPC™ PlugHarvestor™

dPC™ gel plugs must be removed from the chip before in-gel tryptic digestion of proteins or peptide elution before mass spec identification. The dPC™ PlugHarvestor™ allows users to select and group any set of pH gel plugs into the same tube or microplate.

The dPC™ PlugHarvestor™ removes plugs from the dPC™ chip using both physical displacement and hydraulic force in an easy and reliable way. Gel plugs can be pooled together allowing researchers the ability to determine custom fraction sizes. Collection of individual gel plugs allow the researcher to collect fractions as small as 0.05 pH units.
